Abstract

Plastics have become an integral part of our modern society, largely because they are generally regarded as chemically inert under normal circumstances. However, plastics fragmented into microplastics (less than 5 mm in size) have recently raised significant public concerns, and have been detected widely across ecosystems and human tissues with increasing environmental and health risks1–5. Here we show that commodity microplastics exhibit intrinsic physicochemical activity in water, manifested by the spontaneous generation of reactive oxygen species that increases with interfacial hydrophobicity and strongly correlates with interfacial electrostatics. Ab initio calculations support these observations and reveal that hydroxide oxidation becomes energetically more favourable at increasingly hydrophobic microplastic–water interfaces through the combined effects of partial solvation and electrostatics. These interfacial properties and the resulting oxidative chemistry have direct implications for biomolecular adsorption, protein aggregation, and even cellular damage. Together, our findings challenge the prevailing paradigm of plastic inertness, identify a possibly overlooked mechanism for the detrimental effects of microplastics on health and the environment, and contribute to a fundamental understanding of the structure–activity relationship of interfacial water, which may inform the design of safer and more sustainable plastic materials. Commodity microplastics exhibit intrinsic physicochemical activity at water interfaces, manifested by spontaneous generation of reactive oxygen species, challenging the prevailing paradigm of plastic inertness, with implications for human health and the environment.

Strengths · Limitations

Strengths

#methodological rigour

Complementary assays converge on interfacial ROS

Methyl-orange degradation, DMPO ESR, spatially resolved APF imaging, and HPLC conversion measurements provide different views of the observed chemical activity. Solvent and surfactant perturbations further connect the signals to the aqueous interface.

↳ Fig. 1; Methods, ESR measurements and APF radical sensing assay

#contribution

A mechanistic bridge across polymer interfaces

Comparisons across six commodity polymers connect measured reactivity with hydrophobicity, while calculations provide a partial-solvation and electrostatic explanation for relative energetic trends.

↳ Figs. 2–3; Chemical activity at aqueous microplastics surface; Lower vertical ionization energies

#methodological rigour

Computational assumptions receive explicit sensitivity checks

The modelling account includes force-field benchmarking, a polymer-inclusion sensitivity check, and trajectory-window comparisons. It also states that complete electron removal retains an approximately 100 kcal/mol energetic cost.

↳ Methods, Further benchmarking of force-field combinations and VIE calculations and sampling details; Fig. 3

Limitations

#methodological rigour

Independent replication units remain incompletely specified

The Methods report technical wells, imaging fields, and particle-level measurements without consistently defining independent preparation or animal counts. Correlations and the caffeine suppression claim lack sufficient inferential reporting to establish precision.

↳ Methods, Methyl orange degradation, APF radical sensing assay, ThT assay, and Haemolytic assay

#positioning

Intrinsic attribution leaves alternative pathways unresolved

Stirring and vortexing are not separated from possible mechanically driven electrification, and inert-particle or leachate controls are not reported. The related dielectric-powder contact-electro-catalysis precedent is not substantively distinguished from the proposed intrinsic pathway.

↳ Methods, Methyl orange degradation and APF radical sensing assay; Discussion; ref. 31

#reporting

Biological implications exceed demonstrated exposure relevance

Model protein and erythrocyte assays at high particle loadings do not establish damaging activity at physiological exposures. Haemolysis is associated with interfacial activity across polymers but is not tied to ROS by a scavenger-rescue experiment.

Nabu Commentary

The complementary assays in Fig. 1 support ROS activity in the tested microplastic suspensions, and Figs. 2–3 provide a substantive structure–activity hypothesis. Independent preparation and animal counts are not consistently stated, limiting interpretation of precision and reproducibility. The intrinsic framing also requires stronger separation from leachate effects and mechanically driven electrification, particularly given the stirring protocol and related dielectric-powder literature. The biological assays establish model-system responses rather than the physiological relevance implied by the abstract and Discussion.

Convergent ROS assays and interfacial modelling meaningfully connect commodity-polymer surface properties with aqueous reactivity. The advance remains incompletely distinguished from dielectric-powder contact-electro-catalysis, and intrinsic attribution is not fully isolated.

Methodological Rigour
3.3

Orthogonal assays, solvent and surfactant perturbations, vendor comparisons, and computational checks support the experimental program. Missing independent-unit counts, small cross-condition correlations, and unresolved attribution controls introduce meaningful uncertainty.

3.3

The observation-to-mechanism sequence is followable and the remaining electron-removal cost is acknowledged. Direct biological implications and physiologically relevant rates are asserted more strongly than the model-system evidence establishes.

Positioning
2.8

The paper engages interfacial-water and microplastic-toxicology literature and describes computational approximations. It does not adequately address mechanical activation and additive/leachate alternatives to intrinsic reactivity, while environmental qualifications do not constrain the physiological extrapolation.
